The Science

The 12 Hallmarks of Aging Framework

In 2013, López-Otín and colleagues published a landmark framework identifying nine hallmarks of aging in Cell. The framework was updated and expanded to 12 hallmarks in 2023 (López-Otín et al., Cell, 2023), adding Disabled Macroautophagy, Chronic Inflammation, and Dysbiosis as new pillars. Today this is the dominant scientific framework for understanding the molecular and cellular drivers of biological aging. Each hallmark represents a distinct but interconnected mechanism — understanding which are most active in your biology is the first step toward targeted intervention.
